Excess DDQ (2,3-dichloro-5,6-dicyano quinone) is dissolved in DMSO or DMF, and added to the solution of leuco malachite green – target conjugate (i.e oligonucleotide previously modified with leuco-malachite green nhs ester).  The leuco-malachite green modification group is immediately oxidized to malachite green, which results in an intense green-blue coloration.  The resulting malachite green – target conjugate can be purified by an appropriate method.
