Genomic DNA Isolation Using Buccal (Mouth) Swabs

      
In the buccal swab collection method for genomic DNA isolation, individuals gently swab their inner mouth to collect cells for DNA extraction.  Swabs can be stored at ambient temperature prior to genomic DNA extraction.  PCR primers are designed which bracket the region of interest to detect and analyze continuous segments of genomic DNA.  PCR amplification is then employed to generate double stranded DNA strands that can be employed for direct DNA sequencing, or for resolution of short tandem repeats (STRs) in forensics, paternity and ancestry analysis.
     

 

Method Synopsis:  Two-Step Genomic DNA IsolationTM Kit

After swabbing the inner mouth, the individual inserts the swab in packaging to prevent further DNA contamination.  The swab can be delivered in the mail to the processing laboratory if genomic DNA isolation procedures are done remotely:  Genomic DNA is stable over an extended mailing period. 

Once the swab reaches the processing laboratory, the swab is cut or removed from its handle and placed in the "Genomic DNA Isolation Solution 1 (GDIS-1)" in a DNA-free 1.5 ml tube and heated to 95oC for 5 minutes.  The solution is cooled on ice and then the provided "Genomic DNA Isolation Solution 2 (GDIS-2)" is added to the tube.  The swab is removed using a forcep that has been flamed (or sterilized) to eliminate contaminating DNA.  The resulting solution, which contains isolated genomic DNA, can be stored at -20oC for an extended time period prior to PCR amplification.  This genomic DNA isolation method provides 720 µl of extracted DNA solution per swab; however, only 5-10 µl of this solution is required for PCR amplification.  Therefore, a substantial number of PCR reactions can be performed from the extracted genomic DNA sample.

The entire genomic DNA isolation process requires less than 10 minutes and is performed in essentially two steps, which is a substantially more efficient and rapid method than commercially available kits that employ spin column isolation.  The absence of blood draw and the ease of genomic DNA isolation provided by the FIVEphoton Biochemicals Two-Step Genomic Isolation Kit facilitates wide scale genetic analysis for academic and commercial researchers, and for field analysis.
