Human Soluble alpha-Klotho (SAKL) ELISA Kit (96T)

   Protocol

Introduction

Membrane bound Klotho is 130kDa type-I membrane protein that is related to β-glucuronidases.  Shedding of the ectodomain results in the generation of 70kDa soluble alpha-klotho fragment detected by this ELISA kit.  Reduced production of this protein has been observed in patients with chronic renal failure (CRF), and this may be one of the factors underlying the degenerative processes (e.g., arteriosclerosis, osteoporosis, and skin atrophy) seen in CRF. Also, mutations within this protein have been associated with ageing, bone loss and alcohol consumption. Transgenic mice that overexpress Klotho live longer than wild-type mice.  Klotho has been characterized as modulating amyloid beta pathogenesis in Alzheimers disease.

Recovery

Matrices listed below were spiked with certain level of and the recovery rates were calculated by comparing the measured value to the expected amount of in samples.

Matrix Recovery range(%) Average(%)
serum(n=5) 85-101 92
EDTA plasma(n=5) 91-100 98
heparin plasma(n=5) 86-103 94
Linearity

The linearity of the kit was assayed by testing samples spiked with appropriate concentration of and their serial dilutions. The results were demonstrated by the percentage of calculated concentration to the expected.

Sample 1:2 1:4 1:8 1:16
serum(n=5) 87-104% 89-105% 85-105% 92-104%
EDTA plasma(n=5) 83-91% 86-100% 82-99% 87-97%
heparin plasma(n=5) 86-98% 80-97% 80-98% 80-94%
CV(%)

Intra-Assay: CV<8%
Inter-Assay: CV<10%
