Intoduction to Sumo  1

SUMO 1 functions in a manner similar to ubiquitin in that it is bound to target proteins as part of a post-translational modification system.  SUMO1 is involved in a variety of cellular processes, including nuclear transport, transcriptional regulation, apoptosis, and protein stability.  SUMO1 becomes functionally active when the last four amino acids of the carboxy-terminus are cleaved.  SUMO 1 is also associated with SERCA2A, the sarco/endoplasmic reticulum Ca2+ ATPase that regulates intracellular calcium levels.  SUMO 1 activates and stabilizes SERCA2A by binding  to lysine resides 480 and 585.   This ELISA kit detects free SUMO1 and conjugated SUMO1.

Methods
Overview

The included ELISA micro-strip plate is provided pre-coated with an a monoclonal capture antibody selective to the antigen.  Samples are pre-mixed with a biotin-linked detection antibody.   Chromogen solutions are added, resulting in a reaction with HRP that yields a blue colored solution.  A stop solution is applied to terminate the reaction, and the solution is measured at 450nm.

   Sandwich ELISA Methods to SUMO1

ELISA Assay Steps
  1. Mix sample, provided biotinylated-detection antibody and provided streptavidin-HRP.
  2. Apply sample and antibody-HRP complex to ELISA plate for 60min at 37oC.  Wash 5 times.
  3. Apply chromogens, incubate, then stop reaction.
