Synopsis of ELISA Protocol

Mix samples with the provided biotinylated-detection antibody and streptavidin-HRP solutions.
Apply mixture to ELISA plate for 60min at 37^oC. Then wash 5 times.
Apply and incubate with chromogens, stop the reaction and measure at 450nm.

Sandwich ELISA Method Overview

The included 96-well ELISA micro-strip plate is provided pre-coated with an affinity purified monoclonal antibody selective to the antigen. The sample, an affinity purified polyclonal biotinylated antibody and streptavidin-HRP are added together to the plate and incubated for 60 min at 37^oC. Unbound antibody is washed away, and color reagents are added, which upon conversion by HRP, become blue colored. A stop solution is added causing the solution to become yellow. An absorbance multiplate reader is used to quantitate the colorimetric reaction at 450 nm.

Background on Protein Targets of the Amyloid ELISA Kits:

Amyloid-beta (Abeta) is generated by sequential cleavage of the amyloid precursor protein (APP) by beta and gamma secretases. Beta-secretase cleaves an extracellular fragment of APP and releases a soluble fragment, whereas gamma secretase cleaves within the transmembrane domain and generates amyloid-beta (Aβ) isoforms of 36-43 amino acid residues in length. The dominant isoforms of Amyloid-Beta are Aβ₄₀ and Aβ₄₂. Aβ₄₀ is the most common amyloid-Beta (Aβ) isoform, whereas Aβ₄₂ is the more fibrillogenic and the predominant Abeta peptide associated with Alzheimer’s disease. Antibodies employed in this amyloid-beta ELISA kit are selective to the Aβ₄₂isoform of amyloid-beta, with insignificant cross-reactivity to the Aβ₄₀isoform.
