ELISA Kits (2X96T)
FIVEphoton Biochemicals offers high sensitivity ELISA kits for pathways in cell signaling research. Please use the Available Options menu above to select the kit for ordering.
These kits employ the sandwich ELISA method. Each kit is for 96 determinations, and uses HRP conjugated polyclonal antibody with chromogen A and B for detection.
(ELISA kits are shipped 2-3 weeks after the order. A representative manual is displayed here. Specific manuals are shipped with the kits, or can be obtained by contacting technicalsupport@fivephoton.com).

Steps of Sandwich ELISA

4. Add the color reagent, incubate 15 min, wash

Sandwich ELISA Method Overview
The provided ELISA plate has an antibody selective to the antigen bound to a 96-well dish. Upon addition of samples to the wells, the antigen in solution binds to the antibody and becomes indirectly linked to the solid support. After washes of unbound materials, a second polyclonal antibody conjugated with horse-radish peroxidase (HRP) is added, followed by a second incubation period. Unbound antibody is washed, and color reagents are added, which upon conversion by HRP, become blue colored. A stop solution is then added to block further reaction between HRP and the colorimetric substrates, causing the solution to become yellow. An absorbance multiplate reader is used to quantitate the colorimetric reaction.
These ELISA kits provide a standard peptide solution with known concentration to calibrate absorbance readings to concentration.
