Introduction:

TTR misfolding and aggregation is known to be associated with thee amyloid diseases senile systemic amyloidosis (SSA), familial amyloid polyneuropathy (FAP), and familial amyloid cardiomyopathy (FAC).  Transthyretin tetramer dissociation is known to be rate-limiting for amyloid fibril formation. However, the monomer also must partially denature in order for TTR to be mis-assembly competent, leading to a variety of aggregate structures, including amyloid fibrils.  TTR is also thought to have beneficial side effects, by binding to beta-amyloid protein, thereby preventing beta-amyloid's accumulation into the plaques associated with the early stages of Alzheimer's Disease.

Protocol Summary

1.             Wash plate 2 times before adding standard, sample and control (zero) wells!
2.          Add 100μL standard or sample to each well and incubate for 90 minutes at 37°C
3.          Aspirate and wash plates 2 times
4.          Add 100μL Biotin-labeled antibody working solution to each well and incubate for 60 minutes at 37°C
5.          Add 100 μLSABC Working Solution into each well and incubate for 30 minutes at 37°C
6.          Aspirate and wash plates 5 times
7.          Add 90μL TMB Substrate. Incubate 15 -30 minutes at 37°C
8.          Add 50μL Stop Solution. Read at 450nm immediately
9.          Calculation
