Background on trkA
trkA is a protein encoded by the NTRK1 gene. The trkA kinase is a high affinity membrane bound receptor for Nerve Growth Factor, that phosphorylates itself and members of the MAPK pathway when a neurotrophin binds. It's presence leads to cell differentiation and specificity of sensory neuron subtypes. Mutations of this gene lead to self-mutilating behavior, mental retardation, and cancer. 


Methods Overview:  Sandwich ELISA
This rat trkA ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for trkA has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for trkA is added subsequently and then followed by washes with PBS or TBS buffer.  Avidin-Biotin-Peroxidase Complex is added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of yellow solution is proportional to the amount of rat trkA captured on the plate.

   Representative Sandwich ELISA Methods    

  1. Add sample to plate, incubate 90 min, 37oC.
  2. Add detection antibody, incubate 1 hr, 37oC.
  3. Add ABC solution, incubate 30 min 37oC.
  4. Add chromogens, incubate in dark for 25 min.
  5. Add stop solution.  Measure OD at 450nm.
