Amyloid-Beta N3pE-42 ELISA Kit (Human Specificity)
Sensitive and selective colorimetric ELISA kit to ABeta N3pE-42 with assay range 7.75-496 pg/ml. Highly selective to amyloid-beta N3pE-42 with minimal cross reactivity to unmodified amyloid-beta 1-40 and amyloid-beta 1-42 (see data below). The affinity purified monoclonal antibody absorbed on the plate and the affinity purified polyclonal-HRP detection antibody were generated to ABeta N3pE-42.

Background on Amyloid-Beta N3pE-42
Amyloid-beta (Abeta) is generated through amyloid precursor protein (APP) cleavage by certain secretases. Beta-secretase cleaves an extracellular portion of APP and frees a soluble peptide. Gamma-secretase cleaves APP within the transmembrane domain, creating amyloid-beta (Aβ) isoforms with lengths between 36-43 amino acid residues. In N3pE amyloid beta isoforms, the 3rd glutmate residue is converted pyroglutamate. N3pE-42 and N3pE-40 variants have been detected that appear to have a higher propensity toward aggregation than unmodified amyloid beta, which has been hypothesized to contribute to the Alzheimer's pathology. This kits selectively detects the N3pE-42 amyloid beta variant, whereas a separate ELISA Kit (Part No. 27418) has been developed to detect the N3pE-40 variant. The affinity purified monoclonal antibody percolated on the ELISA plate and the affiniy purified polyclonal HRP conjugated detection antibody in this kit are selective to AβN3pE-42. The antibodies have been generated to the N-terminus of AβN3pE-42.
