Calcium Phosphate Transfection Kit
Calcium phosphate transfection is a simple, time efficient, and relatively inexpensive method to overexpress plasmid DNA in easy to transfect cells such as HEK293T, CHO and BHK cells. The calcium phosphate transfection technique requires few manipulative steps and maintains high levels of reproducibility from experiment to experiment. The FIVEphoton Biochemicals Calcium Phosphate Transfection Kit provides pre-configured reagents and an instructive, in-depth protocol manual that details the steps of calcium phosphate transfection.
Characteristics of Calcium Phosphate Transfection
Calcium phosphate transfection method enables gene introduction into cells in the presence of serum and antibiotics. The calcium phosphate transfection solution can be kept overnight on cells without requiring a change in media on the same day as transfection. In contrast to the calcium phosphate transfection method, liposome or cationic transfection methods are typically cytotoxic to cells during overnight exposure, which requires the researcher to remove the transfection solution several hours after transfection.
In preparation for calcium phosphate transfection, DNA is introduced into cells by subcloning into an expression vector with a CMV driven promotor. The gene encoded by the cDNA that is inserted in the expression vector is subsequently transcribed and translated into a protein. Calcium phosphate transfection allows for the simultaneous introduction of multiple plasmid DNAs into cells.
The calcium phosphate transfection method can often be applied to detect expression of introduced genes after an overnight transfection period, although a 48 hr transfection period is optimal. The image of cells displayed by clicking the Data and Images tab was taken after an overnight (16 hr) transfection period of plasmid DNA encoding red fluorescent protein in HEK293T cells.
Suitable Cell Types for Calcium Phosphate Transfection
The Calcium Phosphate Transfection technique is ideal for introduction of cDNAs into HEK293T, HEK293, CHO and BHK cells, and has been reported to operate effectively in more highly differentiated cell lines, including neuronal cell lines. However, FIVEphoton Biochemicals recommends that the researcher restrict use of calcium phosphate transfection methods to HEK293, HEK293T, CHO and BHK cells.
