Background
 
Members of the ADAM family are transmembrane proteins with a unique structure possessing both potential adhesion and protease domains. Sheddase, a generic name for the ADAM metallopeptidase, functions primarily to cleave membrane proteins at the cellular surface. Once cleaved, the sheddases release soluble ectodomains with an altered location and function.
 
Although a single sheddase may “shed” a variety of substances, multiple sheddases can cleave the same substrate resulting in different consequences.  In neurons, ADAM10 is the most important sheddase with α-secretase activity for proteolytic processing of the amyloid precursor protein.

Protocol Summary

1.         Wash plate 2 times before adding standard, sample and control (zero) wells 
2.          Add 100μL standard or sample to each well and incubate for 90 minutes at 37°C
3.          Aspirate and wash plates 2 times
4.          Add 100μL Biotin-labeled antibody working solution to each well and incubate for 60 minutes at 37°C
5.          Add 100 μLSABC Working Solution into each well and incubate for 30 minutes at 37°C
6.          Aspirate and wash plates 5 times
7.          Add 90μL TMB Substrate. Incubate 15 -30 minutes at 37°C
8.          Add 50μL Stop Solution. Read at 450nm immediately
9.          Calculation
