Mouse Soluble Amyloid Precursor Protein Beta (sAPPβ) ELISA Kit 

Sensitive and selective colorimetric ELISA kit to sAPPβ with assay range 4.1 - 260 pg/ml.  Highly selective to sAPPβ with minimal cross reactivity to sAPPα.  96T

Specifications:
  1. Complete 96T ELISA kit in sandwich format.  Colorimetric detection at 450 nm.
  2. ELISA plate is coated with an affinity purified polyclonal capture antibody that recognizes mouse sAPPβ.  The detection antibody is also a polyclonal antibody that recognizes mouse sAPPβ at its opposite terminus.
  3. The kit has been validated to detect sAPPβ in EDTA-plasma and cell culture media.
  4. EIA buffer contains 1% BSA, 0.05% Tween 20 in PBS
  5. Incubation times:  Samples with plate:  overnight.  Detection antibody:  1 hr
  6. Assay range:  13 pg/ml - 800 pg/ml
  7. Sensitivity:  2.87 pg/ml

Background on sAPPbeta

sAPPbeta is the extracellular soluble fragment of amyloid precursor protein (APP) cleaved by beta-secretase into a 654 amino acid peptide. Cleavage of APP by beta-secretase in the extracellular domain is the first cleavage processing step generating amyloid-beta peptide.  Membrane embedded gama-secretase subsequently cleaves APP within the transmembrane domain and generates amyloid-beta peptide of variable lengths.1 This sAPP beta ELISA detects the sAPP beta peptide in cell culture medium and plasma.





Methods Overview
  1. Equilibrate plate to RT
  2. Add sample to plate, incubate overnight
  3. Add detection antibody, incubate 1 hr
  4. Add color reagents, 30 min
  5. Add stop solution.  Measure OD at 450nm




Storage:  4oC, 1 year
Safety:  Stop solution contains acid. Avoid contact, wear eye protection.
