Introduction

The protein tau is crucial for stabilizing microtubules within neurons, which are essential for their structure and function. Tau can undergo  phosphorylation, where phosphate groups are added to it at various sites. This modification can change tau's shape, its ability to bind to microtubules, and its interactions with other proteins. 

• Indicates Alzheimer's Disease Pathology: Elevated levels of p-tau217 in cerebrospinal fluid (CSF) and, importantly, in blood plasma, strongly correlate with the presence of amyloid plaques and tau tangles in the brain – the two hallmark pathologies of AD.  
• Early Detection: Studies suggest that p-tau217 levels rise even in the early stages of AD, often before significant cognitive symptoms appear. This makes it a potential tool for early detection and intervention.  
• Differentiation from Other Dementias: p-tau217 shows a high specificity for AD compared to other neurodegenerative disorders, making it valuable in distinguishing AD from conditions like frontotemporal dementia or Lewy body dementia.  
• Prediction of Disease Progression: Longitudinal studies indicate that the rate of change in plasma p-tau217 levels can predict the progression of AD, including cognitive decline and brain atrophy.

1. Equilibrate plate to RT
   
2. Add sample to plate, incubate overnight
   
3. Add detection antibody, incubate 30 min
   
4. Add color reagents, 30 min
   
5. Add stop solution.  Measure OD at 450nm
