Quinolinic Acid (QA) is a very neurotoxic chemical associated with many brain disorders, including neurodegeneration (alzheimers, ischemia, and amyotrophic lateral sclerosis [ALS]) and psychiatric illnesse like bipolar disorder, depression, and schizophrenia. Quinolinic acid is also correlated with acquired dementia in some AIDS and HIV patients due to the activity of macrophages in the brain. QA is also the biosynthetic precursor to nicotine. Quninolinic Acid is only found in activated microglia and macrophages within the brain where at excessively high levels it acts as an NDMA agonist, causing the neuron to uptake a large amount of calcium which activates apoptosis. Ultimately, Quinolinic Acid causes apoptosis in neurons leading to neurodegeneration.
Methods Overview:
The included ELISA micro-strip plate is provided pre-coated with an a monoclonal capture antibody selective to the antigen. Samples are pre-mixed with a biotin-linked detection antibody and streptavidin-HRP, which are then added to the plate. The plate is washed after a 60 min incubation period. Color reagents are added, resulting in a reaction with HRP that yields a blue colored solution. A stop solution is applied to terminate the reaction, and the solution is measured at 450nm.
