Protocol
Methods Overview:
The included ELISA micro-strip plate is provided pre-coated with an a monoclonal capture antibody selective to the antigen. Samples are pre-mixed with a biotin-linked detection antibody and streptavidin-HRP, which are then added to the plate. A 60 minute incubation period at 37oC is used, and then the plate is subsequently washed. Color reagents are added, resulting in a reaction with HRP that yields a blue colored solution. A stop solution is applied to terminate the reaction, and the now yellow solution is measured at 450nm.

Apply sample and antibody-HRP complex to ELISA plate for 60min at 37oC. Wash 5 times.

Apply chromogens, incubate, then stop reaction.
Introduction to Kallikrein-6:
Kallikrein-6 is a protein with serine protease activity that is detected with grey matter neurons and colocalizes with pathological inclusions. Kallikreins have many known functions; they are associated with carcinogenisis and are thought to be disease biomarkers. Kallikrein-6 has been found to cleave amyloid precursor protein as well as myelin basic protein, gelatin, casein, and many extracellular matrix proteins. Kallikrein-6 also cleaves alpha-synuclein, preventing polymerization.
