Beta-Synuclein (SNCB) is a member of the synucelin protein family.  Beta-synuclein is highly expressed in the substantia nigra, a region of neuronal degeneration in Parkinson's disease patients.  Beta-synuclein may inhibit alpha-synuclein aggregation associated with Parkinson's disease, potentially providing protection from the toxicity associated with alpha-synuclein aggregation.  This ELISA kit is highly selective to beta-synuclein with insignificant cross-reactivity to gamma and alpha-synuclein.


Methods Overview:
The included ELISA micro-strip plate is provided pre-coated with an a monoclonal capture antibody highly selective to SNCB.  Samples are pre-mixed with a biotin-linked polyclonal detection antibody specific to beta-synuclein and streptavidin-HRP;  the mixture is then added to the plate and incubated for 60 min.  The plate is washed after the incubation period.  Color reagents are added, resulting in a reaction with HRP that yields a blue colored solution.  A stop solution is applied to terminate the reaction.  The solution is measured at 450nm.

Rapid Protocol ELISA Assay Steps
 
  1. Mix sample, biotinylated-detection antibody and streptavidin-HRP.
  2. Apply sample and antibody-HRP complex to ELISA plate for 60min at 37oC.  Afterwards, wash 5 times.
  3. Apply chromogens, incubate, then stop the reaction.
