TMPRSS2 is a type II transmembrane protein thought to be cleaved and secreted into cell media after autocleavage. TMPRSS2 is involved in viral entry by cleaving surface glycoproteins and priming for infection. Viruses found to use this serine protease for cell entry include Influenza virus and the human coronaviruses HCoV-229E, MERS-CoV, SARS-CoV and SARS-CoV-2 (COVID-19). TMPRSS2 mediated cleavage in the S2 domain appears to be modulate the efficiency of SARS-CoV2 infection. TMPRSS2 facilitates entry of viruses into host cells by proteolytically cleaving and activating viral envelope glycoproteins.
Step 1: Wash plate twice before adding Standard, Samples (diluted at least 1/2 with Sample Dilution Buffer) and Control (blank) wells.
Step 2: Add 100ul standard or sample to each well and incubate for 90 minutes at 37°C. Wash step: Aspirate and wash plates 2 times.
Step 3: Add 100ul Biotin-labeled antibody working solution to each well and incubate for 60 minutes at 37°C. Wash step: Aspirate and wash plates 3 times.
Step 4: Add 100ul SABC Working Solution into each well and incubate for 30 minutes at 37°C. Wash step: Aspirate and wash plates 5 times.
Step 5: Add 90ul TMB Substrate Solution. Incubate 10-20 minutes at 37°C.
Step 6: Add 50ul Stop Solution. Read at 450nm immediately and calculation.
