Background on Protein Targets of the Amyloid ELISA Kits

Amyloid-beta (Abeta) is generated by sequential cleavage of the amyloid precursor protein (APP) by beta and gamma secretases.  Beta-secretase cleaves an extracellular fragment of APP and releases a soluble fragment, whereas gamma secretase cleaves within the transmembrane domain and generates amyloid-beta (Aβ) isoforms of 36-43 amino acid residues in length.  The dominant isoforms of Amyloid-Beta are Aβ₄₀ and Aβ₄₂.   Aβ₄₀ is the most common amyloid-Beta (Aβ) isoform, whereas Aβ₄₂ is the more fibrillogenic and the predominant Abeta peptide associated with Alzheimer’s disease.  This ELISA kit is highly selective to amyloid beta 1-40 with insignificant cross-reactivity to amyloid beta 1-42.  The kit detects the full length amyloid beta 1-40 peptide using capture and detection monoclonal antibodies that recognize epitopes at the 1 and 40 positions.
