Soluble Amyloid Precursor Protein Beta wild type (sAPPβ-w)ELISA Kit (Human Specificity;  96T)  

Sensitive and selective colorimetric ELISA kit to sAPPβ-w with assay range 0.78-50 ng/ml.  Highly selective to sAPPβ-w with minimal cross reactivity to sAPPalpha and sAPPβ-sw (see data below).  The affinity purified polyclonal capture antibody recognizes the C-terminus of the sAPPbeta cleavage product by Beta-secretase and the monoclonal detection antibody recognizes an epitope on the N-terminus of sAPPbeta.

Specifications:
  1. Complete 96T ELISA kit in sandwich format.  Colorimetric detection at 450 nm.
  2. ELISA plate is coated with an affinity purified polyclonal antibody that recognizes the C-terminus sAPPβ-w.  The detection antibody is a monoclonal that binds to an epitope on the N-terminus.
  3. The kit detects sAPPβ-w in EDTA-plasma, cerebrospinal fluids, cell culture media or brain tissue extract.

Background on sAPPβ

When beta-secretase cleaves the extracellular domain of amyloid precusor protein (APP) in the first cleavage-processing step towards generating amyloid-beta peptide, the extracellular soluble fragment is known as sAPPbeta. In another cleavage-processing step, gama-secretase embedded in the membrane subsequently cleaves APP within the transmembrane domain and forms amyloid-beta peptide of variable length.1 This sAPP beta ELISA detects the sAPP beta peptide in cell culture medium and biological fluids, including CSF.


Methods Overview
  1. Equilibrate plate to RT
  2. Add sample to plate, incubate overnight
  3. Add detection antibody, incubate 1 hr
  4. Add color reagents, 30 min
  5. Add stop solution.  Measure OD at 450nm
