Sensitive and selective colorimetric ELISA kit to ABeta Oligomers with assay range 18.98-1215 pmol/L.  Highly selective to amyloid-beta oligomers with minimal cross reactivity to amyloid-beta 1-40 peptide (see data below).  The affinity purified monoclonal antibody absorbed on the plate and the affinity purified polyclonal-HRP detection antibody are designed to operate as a pair to detect ABeta oligomers.

Background on Amyloid-Beta Oligomers

Amyloid-beta (Abeta) is generated through amyloid precursor protein (APP) cleavage by certain secretases.  Beta-secretase cleaves an extracellular portion of APP and frees a soluble portion. Gamma-secretase cleaves APP within the transmembrane domain, creating amyloid-beta (Aβ) isoforms with lengths between 36-43 amino acid residues.  The prevailing isoforms of Amyloid-Beta are Aβ40 and Aβ42.  Aβ40 is the most widespread amyloid-Beta (Aβ) isoform, while Aβ42 is more fibrillogenic and is the principal Abeta peptide associated with Alzheimer’s disease.  The affinity purified monoclonal antibody percolated on the ELISA plate and the affiniy purified polyclonal HRP conjugated detection antibody in this kit operate as a pair to detect amyloid-beta oligomers.

Methods Overview
  1. Equilibrate plate to RT
  2. Add sample to plate, incubate overnight
  3. Add detection antibody, incubate 1 hr
  4. Add color reagents, 30 min
  5. Add stop solution.  Measure OD at 450nm
