Protocol Synopsis for Fivephoton Cell Viability and Proliferation Assay

  1. Plate cells in 96-wells dishes and culture until approximately 85% confluency.   (Other mutliwell dishes are acceptable as well).  Use "control"  cells for baseline comparison.
  2. Add WST-8 reagent.
  3. Incubate cells 1-4 hours.
  4. Measure OD at 450 nm.
  5. The relative absorbance at 450 nm provides an index of viable cell number.

Reaction scheme involving NADPH and conversion of colorless WST-8 into its orange reduced form.

The Fivephoton Cell Viability and Proliferation Assay Kit is a colorimetric assay for determining the number of viable cells in proliferation or cytotoxicity assays that uses the reagent WST-8 as its substrate. The highly water-soluble tetrazolium salt, WST-8 is cleaved to a soluble formazan by cellular dehydrogenases. This bioreduction is accomplished by NADPH or NADH produced by dehydrogenase enzymes in metabolically active cells. The amount of formazan dye product directly correlates to the number of metabolically active cells in the culture.  Because the WST-8 formazan dye is soluble in tissue culture medium, the Fivephoton Cell Viability and Proliferation Assay Kit requires fewer steps than procedures that use the tetrazolium compound MTT.  The results  obtained  by using  the  kit  show  higher  sensitivity  than  methods  that  use tetrazolium compounds such as MTT, MTS, XTT or WST-1.

The ready-to-use WST-8 solution does not require additional steps such as thawing before use.  Therefore, the storage stability of the Fivephoton Cell Viability and Proliferation kit is higher than kits using other tetrazolium compounds.
